Understanding Metformin and its Absorption

Metformin’s absorption primarily occurs in the small intestine. About 50-60% of an oral dose is absorbed, with peak plasma concentrations reached within 2-3 hours. Food significantly impacts absorption; taking metformin with food reduces the rate and extent of absorption, leading to lower peak levels.

Factors Influencing Absorption

Several factors influence how well your body absorbs metformin. These include gastric emptying rate (faster emptying leads to quicker absorption), intestinal transit time (faster transit reduces absorption), and the presence of other medications. For instance, some drugs can interact and affect metformin’s absorption.

Renal function plays a critical role. Metformin is primarily excreted unchanged by the kidneys. Reduced kidney function significantly decreases metformin clearance, potentially leading to higher blood levels and a greater risk of side effects. Regular monitoring of kidney function is therefore necessary, especially for individuals at increased risk or with pre-existing renal conditions.

Individual variations also exist. Genetic factors and individual differences in gut microbiota can influence the absorption process. These factors explain why some people experience different absorption rates than others.
