Mechanism of Action: How Amoxicillin Works

Amoxicillin inhibits bacterial cell wall synthesis. It achieves this by binding to penicillin-binding proteins (PBPs), enzymes crucial for peptidoglycan formation. Peptidoglycan is a vital component of the bacterial cell wall, providing structural integrity.

Inhibition of Transpeptidases

Amoxicillin specifically targets bacterial transpeptidases, a subset of PBPs. These enzymes catalyze the cross-linking of peptidoglycan strands, creating a strong, rigid cell wall. By blocking transpeptidase activity, amoxicillin prevents this cross-linking.

Consequences of Inhibition

The weakened cell wall renders bacteria susceptible to osmotic lysis. Cellular pressure from within the bacterium overcomes the compromised cell wall’s ability to withstand it, leading to cell death. This mechanism is most effective against actively growing bacteria.
